HEART DAMAGE WITH BACLOFEN (EXPERIMENTAL STUDY)

Abstract

Objective. To identify functional and metabolic disorders of the heart under the influence of toxic doses of baclofen.Material and methods. Two series of acute experiments were carried out on male Wistar rats weighing 200-300 g. Animals of the control series received 0,9% sodium chloride solution 6 ml/kg of body weight, whereas experimental group animals received baclofen at a dose of 145 mg/kg of body weight in normal saline solution through a gastric tube. General inhalation anesthesia induced using sevoflurane 4 vol% with an oxygen flow of 2 L/min in an induction chamber. The animals positioned supine on operating table. Inhaled anesthesia was maintained through the mask and anesthesia machine on spontaneous breathing. ECG, heart rate (min-1), respiration rate (RR, ЭКСПЕРИМЕНТАЛЬНОЕИССЛЕДОВАНИЕ     33Shoshilinch tibbiyot axborotnomasi, 2020, 13-tom, 5-sonmin-1) were recorded, blood pressure was invasively measured with a BP-100 device. Arterial blood samples were taken for biochemical studies. Euthanasia was carried out by intra-arterial injection of 2% lidocaine solution under general anesthesia with chloral hydrate. Samples of the myocardium were fixated for morphological examination.Results. Baclofen in toxic doses disrupted the function of automatism and conduction, caused bradypnea. Morphologically, microcirculation disturbances such as plasma separation, stasis, sludge and microthrombosis, perivascular edema and hemorrhages, as well as contractures of cardiomyocytes, their wavy deformation and nuclear hyperchromia are revealed in the heart muscle.Key words: baclofen, morphofunctional heart damage.
